Lipids rewire T cell exhaustion
Nature Immunology, Published online: 27 June 2025; doi:10.1038/s41590-025-02177-9
CD8+ T cell exhaustion and dysfunction are features of a number of diseases associated with chronic antigen exposure and in some cases can be overcome with checkpoint-blocking therapies. Researchers are now investigating the crosstalk between cellular exhaustion and metabolism with a focus on the role of lipid mediators and redox biology.
Summary
A Nature Immunology article (June 2025) explores the interplay between CD8+ T cell exhaustion, a hallmark of chronic antigen exposure in diseases, and cellular metabolism. The study focuses on the influence of lipid mediators and redox biology in driving T cell dysfunction, a state that can sometimes be reversed by checkpoint inhibitors. Researchers are aiming to understand how metabolic processes contribute to T cell exhaustion, potentially revealing new therapeutic targets for enhancing immune responses in these challenging conditions.
