Potentiating cancer immunotherapies with modular albumin-hitchhiking nanobody–STING agonist conjugates

This research explores a novel approach to enhance cancer immunotherapy by delivering STING (Stimulator of Interferon Genes) agonists directly to tumors using albumin-hitchhiking nanobody conjugates. The conjugates consist of a tumor-targeting nanobody, a STING agonist, and a modified albumin-binding domain (ABD). The ABD allows the conjugate to bind to albumin in the bloodstream, extending its circulation time and enhancing tumor accumulation. This targeted delivery of STING agonists activates the STING pathway specifically within the tumor microenvironment, promoting anti-tumor immune responses and potentially improving the efficacy of cancer immunotherapies while minimizing systemic side effects.
