This study investigates the relationship between the functional avidity of anti-B7H3 CAR-T cells and the antigen density required for triggering effector function. By analyzing various CAR constructs with different binding affinities to B7H3, the research demonstrates that CAR-T cells with higher functional avidity can be activated by lower levels of B7H3 antigen expression. This suggests a crucial role for functional avidity in determining the sensitivity of CAR-T cells to tumor cells expressing varying levels of B7H3. The findings emphasize the importance of optimizing CAR design for improved efficacy against tumors with low antigen density.
