Metabolic reprogramming via FOXP3 engineering: A Novel strategy to enhance CAR-T cell efficacy in solid tumors

Engineering CAR-T cells with the transcription factor FOXP3 can reprogram their metabolism to improve effectiveness against solid tumors. Solid tumors often create a nutrient-poor and immunosuppressive environment. By introducing FOXP3, researchers can shift CAR-T cells towards utilizing fatty acid oxidation for energy instead of relying on glucose, which is often scarce in tumors. This metabolic adaptation enhances CAR-T cell survival, persistence, and anti-tumor activity within the challenging tumor microenvironment. This novel approach offers a promising strategy to overcome limitations in CAR-T cell therapy for solid tumors.
