A novel nanoemulsion adjuvant, encapsulating a TLR7/8 agonist, was developed for enhanced and broad-spectrum antiviral immunity. This “Trojan-TLR7/8” adjuvant facilitates efficient antigen delivery and controlled, multiscale immunomodulation. In preclinical studies, this adjuvant, when combined with various viral antigens, elicited strong humoral and cellular immune responses against both endemic and pandemic viruses, even offering protection against heterologous strains. The nanoemulsion promotes sustained immune activation and superior protection compared to traditional adjuvants, highlighting its potential for developing broadly protective vaccines against diverse viral threats.
