Modulating tumor collagen fiber alignment for enhanced lung cancer immunotherapy via inhaled RNA



Summary

Inhaled RNA targeting HSP47, a key collagen chaperone, can modulate tumor collagen fiber alignment and enhance immunotherapy efficacy in lung cancer. By disrupting collagen synthesis and crosslinking, the treatment loosens the dense, aligned collagen network surrounding tumors. This allows for improved T cell infiltration and activation within the tumor microenvironment, crucial for effective immunotherapy. Studies demonstrated that inhaled HSP47-targeting RNA, combined with immune checkpoint inhibitors, significantly improved tumor control and survival rates in preclinical lung cancer models compared to either treatment alone. This approach offers a non-invasive strategy to remodel the tumor microenvironment, potentially boosting immunotherapy responses in lung cancer patients.
