Researchers have developed a directed evolution approach to discover ligands that can specifically restimulate chimeric antigen receptor (CAR) T cells in vivo. By screening a library of peptides expressed on modified MHC molecules in mice, they identified ligands that selectively activate CAR T cells, leading to enhanced tumor control and prolonged survival. This method offers a way to fine-tune CAR T cell activity within the body, potentially overcoming limitations like exhaustion and antigen escape. The approach provides a framework for designing “booster shots” to improve the persistence and efficacy of CAR T cell therapies in treating cancer.
