Pre-TCR-targeted immunotherapy aims to eliminate leukemic T cells in T cell acute lymphoblastic leukemia (T-ALL) by targeting the T cell receptor (TCR) before it fully assembles. This approach overcomes challenges associated with targeting mature TCRs, which can lead to T cell fratricide and antigen escape. Strategies include targeting TCR-associated proteins like CD3 or exploiting unique TCR rearrangements present in T-ALL cells.
