Genetically engineered T cells targeting a specific mutation in the β-catenin protein have shown remarkable success in eradicating solid tumors in mice. This mutation, often found in various cancers, is a promising target due to its presence inside the tumor cells, posing a delivery challenge. Researchers developed T cells engineered with a specific T cell receptor that can recognize and bind to the mutated β-catenin protein when presented on MHC molecules. The study demonstrated that these engineered T cells effectively eliminated tumor cells expressing the mutated protein, leading to complete tumor regression in preclinical mouse models, highlighting the potential of this targeted immunotherapy approach for treating solid tumors.
