Orchestrating intratumoral DC-T cell immunity for enhanced tumor control via radiotherapy-activated TLR7/8 prodrugs in mice
Summary
Researchers developed a novel approach combining radiotherapy (RT) with TLR7/8 agonist prodrugs activated by RT within tumors in mice. This strategy successfully orchestrated an intratumoral immune response, specifically activating dendritic cells (DCs) and T cells, leading to improved tumor control. The RT-activated prodrugs enhanced DC maturation and antigen presentation, which, in turn, stimulated T cell infiltration and cytotoxicity within the tumor microenvironment. This combination therapy outperformed RT alone or non-activated TLR7/8 agonists, demonstrating the potential of locally activating immune stimulants with RT to boost anti-tumor immunity. The study provides a promising strategy for cancer immunotherapy by exploiting RT to trigger targeted immune activation within the tumor.
