The unfolded protein response influences therapy outcome and disease progression in chronic lymphocytic leukaemia



Summary

Chronic lymphocytic leukemia (CLL) cells rely on the unfolded protein response (UPR), a cellular stress response pathway, to survive. The UPR is activated due to protein misfolding and aggregation, a consequence of the high protein production needed for rapid proliferation. High UPR activation in CLL cells is associated with more aggressive disease and poorer response to chemotherapy and targeted therapies like ibrutinib. Conversely, inhibiting the UPR can sensitize CLL cells to these therapies, suggesting it as a potential therapeutic target to improve treatment outcomes and slow disease progression in CLL patients. Targeting the UPR might overcome resistance and improve survival.
