Dual targeting of tumoral cells and immune microenvironment by blocking the IL-33/IL1RL1 pathway
Summary
Blocking the IL-33/IL1RL1 pathway presents a promising dual approach for cancer therapy. IL-33, released by stressed or damaged cancer cells, interacts with its receptor IL1RL1 on both tumor cells and immune cells within the tumor microenvironment. On tumor cells, this interaction can promote proliferation and survival. In the immune microenvironment, IL-33/IL1RL1 can skew immune responses, often promoting immunosuppression. Therefore, inhibiting this pathway can directly target cancer cell growth while simultaneously modulating the tumor microenvironment to enhance anti-tumor immunity, offering a potentially more effective cancer treatment strategy.
