UGCG and the glycosphingolipid rheostat: a metabolic checkpoint governing immune activation and tumor immune evasion



Summary

UDP-glucose ceramide glucosyltransferase (UGCG) controls the “glycosphingolipid rheostat,” a metabolic checkpoint pivotal in immune responses and tumor immune evasion. UGCG initiates the synthesis of complex glycosphingolipids (GSLs). Increased GSLs promote immunosuppression, facilitating tumor growth. Conversely, lower GSLs enhance immune activation. This balance, regulated by UGCG, impacts various immune cell functions and tumor microenvironment composition. Therefore, targeting UGCG presents a therapeutic strategy to modulate the GSL rheostat, potentially boosting anti-tumor immunity or suppressing detrimental inflammatory responses, making it a promising target in cancer immunotherapy and other immune-related diseases.
