Engineering STING Nanoadjuvants for spatiotemporally-tailored innate immunity stimulation and cancer vaccination therapy

Engineering STING-activating nanoadjuvants offers a promising avenue for enhancing cancer immunotherapy by precisely controlling the location and timing of immune stimulation. These nanoadjuvants encapsulate STING agonists, delivering them directly to antigen-presenting cells (APCs) within the tumor microenvironment or draining lymph nodes. This targeted delivery minimizes systemic toxicity and maximizes the activation of innate immunity, particularly the STING pathway. Researchers are tailoring these nanoadjuvants’ properties, such as size, shape, and surface modifications, to optimize cellular uptake and intracellular release of the STING agonist.
